This study was performed to improve the speed of and standardize the process for the germination of macaw palm seeds. The effect of mechanical scarification (removal of the tegument in the hilum region), thermal scarification (imbibition of seeds in water at approximately 100°C for two or four minutes) and chemical scarification (98% sulfuric acid treatment for two or four minutes) were compared to a control (no scarification) in both a germination chamber and a greenhouse, representing trials one and two, respectively. The objective of this study is to establish an in vitro germination and cultivation protocol for murici (Byrsonima cydoniifolia A. Juss.) using zygotic embryos. Therefore, three assays were performed: in assay I, embryo asepsis was tested at exposure times of 5, 10, 15, 20, 25, and 30 minutes in 2.5% sodium hypochlorite, with or without immersion in 70% alcohol; in assay II, MS (MURASHIGE; SKOOG, 1962) e WPM (LLOYD; McCOWN, 1980) culture media were tested at salt concentrations of 25, 50, and 100%, with or without the addition of sucrose, to germinate the buds; in assay III, seedling growth was evaluated in MS and WPM culture media at salt concentrations of 25, 50 and 100%. This study evaluated the effect of light, temperature, physiologic stage at harvest and length of time on the germination of Hyptis marrubioides seeds. Two trials were conducted. The first experiment was performed immediately after the seed harvest and consisted of a 2 x 3 x 3 factorial design with 2 environmental conditions (light and dark), 3 temperatures (20, 30 and 20/30ºC) and 3 seed harvest times (green colored seeds, light brown seeds and dark brown seeds). The influence of moisture on the in vitro embryo germination and morphogenesis of babassu (Orbignya phalerata Mart.) - doi: 10.4025/actasciagron.v34i4.13429 Agronomy

The objective of this study was to evaluate the physiological quality of babassu (Orbignya phalerata Mart.) embryos as a function of desiccation. The fruits were placed in a convection oven at 37±2°C for 0, 3, 6, 9, or 12 days. After these periods, the moisture contents of the fruits and seeds were determined. A reduction in the water content occurred more rapidly in the fruits than in the seeds. The decrease in seed moisture content from 12.59% (on the basis of fresh weight) to 4.56% did not affect the percentage of germinated zygotic embryos; however, we did observe a decline in the vigor as the seed moisture content decreased. Embryos with a 4.56% moisture content developed into seedlings of shorter lengths after 30, 60, and 90 days of in vitro growth....
